What is cell confluency?

Cell confluency is the percentage of the well surface covered by cells, from 0% (empty) to 100% (full monolayer). It is the primary indicator of cell growth status. Most assays require a specific confluency at treatment — typically 70–90% for drug treatment, 100% for scratch assays. Manual estimation by eye introduces 15–25% inter-observer variability. Automated measurement eliminates this entirely.

How does zenCELL owl monitor confluency?

zenCELL owl sits inside the CO₂ incubator and captures brightfield images of all 24 wells at user-defined intervals (5 minutes to 4 hours). Its software automatically segments cell-covered vs. cell-free areas and calculates % confluency per well per timepoint. Data is displayed as a real-time growth curve. Alerts can be set at threshold confluency values — you receive a notification when cells reach 80% without entering the lab.

What vessels are compatible?

zenCELL owl is compatible with 6-, 24-, and 96-well plates, T-25 and T-75 flasks, ibidi chambers, and most standard cell culture formats. It adapts to different vessel sizes via modular plate holders.

Can zenCELL owl monitor confluency in T-flasks?

Yes. zenCELL owl fits inside standard CO₂ incubators and can image T-25 and T-75 flasks alongside multiwell plates. This makes it useful for routine cell culture quality control — tracking growth across multiple flasks simultaneously without opening the incubator.

Why is automated confluency monitoring better than manual estimation?

Manual confluency estimation by eye has inter-observer CV of 15–25% and requires entering the lab and opening the incubator. Automated monitoring provides: objective quantitative data with no observer variability; continuous tracking without incubator disruption; complete growth kinetics (doubling time, lag phase, growth rate); and remote monitoring from outside the lab. What cell lines can be monitored for confluency?

Any adherent cell line is compatible: HeLa, HEK293, CHO, Vero, MCF-7, A549, HUVEC, primary fibroblasts, keratinocytes, MSCs, iPSCs, and all standard research cell lines. Non-adherent suspension cells cannot be measured for confluency.
